Competitive antibody ELISAs work on a slightly different principle. Patient antibodies compete with a biotin-labelled artificial molecule for binding sites on the coated antigen. A peroxidase-labelled avidin conjugate is then introduced, which binds the biotin-labelled molecules. The final TMB reaction produces a color intensity that is inversely proportional to the antibody concentration in the sample, allowing precise quantification.
For antigen detection, sandwich ELISAs are used. Wells are coated with monospecific antibodies that bind to the target antigen in the patient sample. A second peroxidase-labelled antibody binds to another epitope of the same antigen, creating a “sandwich.” Upon addition of TMB, a color reaction occurs, with intensity directly proportional to the antigen concentration.
In competitive antigen ELISAs, patient antigens compete with a biotin-labelled antigen for binding sites on the antibody-coated wells. Peroxidase-labelled streptavidin binds the biotin-labelled antigen, and the final TMB color reaction is inversely proportional to the antigen concentration in the sample. Both approaches allow precise measurement and are tailored for different diagnostic needs.
